show Abstracthide AbstractWe conducted Transposon Directed Insertion-Site Sequencing (TraDIS) on three different strains of Escherichia coli. We determined the number of unique mutants: 1) as a function of recovery time after electroporation (0.5, 1 or 2 hrs); and 2) as a function of selection medium (agar plates or liquid broth). For the recovery analysis, we also compared the recovery of transposon-DNA junctions based on the layout of sequencing library fragments (Nextera-TruSeq hybrid, or TruSeq-TruSeq).
